Temperature Effects on Cholinesterases Rat Brain Capillaries

چکیده

The activities of acetylcholinesterase (ACHE) and butyrylcholinesterase (BuChE) in rat brain capillaries were measured as a function of temperature. Arrhenius plots of the data revealed that AChE exhibits a biphasic Arrhenius plot with a distinct break (transition temperature) at about 15.2 kcal/mol. In contrast, BuChE did not show evidence of discontinuity. BuChE showed an activation energy higher than that of AChE in the physiological range of temperature. These data suggest a lack of lipidprotein interaction in the case of BuChE. Although the possibility exists that BuChE is weakly anchored to the membranes, our results indicate that BuChE is not bound, at least significantly, to cellular membranes in brain capillaries as is ACHE.

Phosalone-Induced Changes in Regional Cholinesterase Activities in Rat Brain during Behavioral Tolerance

Organophosphate pesticides exert their toxic effects by cholinesterase inhibition and the consequent prolongation of the undesirable effects of accumulation of acetylcholine. The signs of toxicity include tremors, convulsions, lachrymation, defecation etc. However, sustained cholinesterase inhibition through sustained administration of organophosphates would lead to the gradual disappearance of...

Tissue cholinesterases. A comparative study of their kinetic properties.

The substrate saturation and temperature-dependent kinetic properties of soluble and membrane-bound forms of acetylcholinestarase (AChE) from brain and butyrylcholinesterase (BChE) from heart and liver were examined. In simultaneous studies these parameters were also measured for AChE in erythrocyte membranes and for BChE in the serum from rat and humans. For both soluble and membrane-bound for...

Optimal dose of plasmid vascular endothelial growth factor for enhancement of angiogenesis in the rat brain ischemia model.

Vascular endothelial growth factor (VEGF) administration has recently been assessed as a therapeutic strategy for ischemic diseases including brain ischemia because of its angiogenic effect. However, VEGF also causes detrimental adverse effects by increasing vascular permeability. This study examined whether plasmid human VEGF (phVEGF) administration induced angiogenic effects in the rat brain ...

Cholinesterases in neural development: new findings and toxicologic implications.

Developing animals are more sensitive than adults to acute cholinergic toxicity from anticholinesterases, including organophosphorus pesticides, when administered in a laboratory setting. It is also possible that these agents adversely affect the process of neural development itself, leading to permanent deficits in the architecture of the central and peripheral nervous systems. Recent observat...
